The test cassette's membrane is pre-coated with anti-hemoglobin antibodies on the test line region, which react with particles coated with anti-hemoglobin antibodies in the specimen. The mixture then migrates upwards on the membrane chromatographically through capillary action to react with anti-hemoglobin antibodies on the membrane, resulting in the generation of a colored line.

The presence of a colored line in the test line region indicates a positive result for the presence of Human Occult Blood in the sample, while the absence of a colored line indicates a negative result. To serve as a procedural control, a colored line always appears in the control line region, indicating that the proper volume of specimen has been added and membrane wicking has occurred.
